dc.description.abstractHUWE1 is an E3 ligase that is involved in ubiquitin mediated degradation and signaling in a variety of cellular processes including DNA damage repair. However, its role in the DNA damage response (DDR) pathway is not well understood. PARP1, is a key protein involved in sensing and initiating DNA damage signaling through catalyzing the attachment of poly-ADP ribose (PAR) polymers to substrate proteins, to mediate the recruitment of various proteins at DNA damage sites. HUWE1 was recently found to bind ADP-ribose through its WWE domain. Therefore, HUWE1 and PARP1 could be associated in the regulation of protein signaling in the DDR pathway. The association of HUWE1 with DDR proteins including PARP1 was studied using biochemical and cellular approaches. HUWE1 was shown to be involved in DNA damage signaling through affecting phosphorylated-ATM and phosphorylated-NBS1 in response to DNA damage. Moreover, HUWE1 was shown to interact with PAR and PARP1 using co-immunoprecipitations.
